STATEMENT OF NEED
Myeloproliferative neoplasms are a heterogeneous group of hematopoietic disorders that includes myelofibrosis, polycythemia vera (PV), and essential thrombocythemia (ET). These malignancies affect approximately 13,000, 134,000, and 148,000 individuals in the United States, respectively. As symptoms do not typically manifest in early stages, patients with MPNs often present with advanced disease. The presence of comorbidities precludes most from potentially curative allogeneic stem cell transplant. Due to the dearth of effective treatment options, therapeutic goals are focused on slowing progression and maintaining quality of life. Median overall survival varies across MPN subtypes, ranging from 15.4 years for patients with low-risk disease to 1.3 years for those with high-risk disease. Furthermore, MPNs are also associated with a risk for transformation to acute myeloid leukemia, where the 5-year overall survival rate is only 25%.
